Site-specific labeling of cellular proteins with unnatural substrates of biotin ligases

In vitro compartmentalization is investigated as a platform for re-engineering the substrate specificity of BirA through in vitro evolution and a new and orthogonal biotin ligase-acceptor peptide pair for site-specific protein labeling is developed.
